Insulin Binding Characteristics of Human Erythrocyte Membranes

Abstract

|
Insulin binding characteristics of isolated human erythrocyte membranes were compared with those of original intact erythrocytes.
The erythrocyte membranes shared the same kinetics and pH optima of insulin binding reaction with the original cells.
Scatchard analysis of the binding data showed the presence of two major classes of binding sites: high-specific and low-specific binding sites. The association constant (Ka) and number of binding sites of high-specific one were found to be 2.7 X 109M-1 and 4 sites/cell in erythrocyte, and 2.3 X 109M-1 and 4 sites/cell in the membrane fraction. The number of low-specific binding sites was slightly decreased in the membrane fraction (40 sites/cell) compared with that of original erythrocytes (53 sites/cell).
These results indicate that the insulin binding of erythrocyte membrane is similar to insulin binding of the original erythrocytes, and suggest that being more stable on storage, erythrocyte membrane may be preferable to whole red cells for long-term insulin receptor studies.
